Untangling the Complexities of Gene Synthesis for Longer, More Reliable DNA

  • Published

At Ansa, we’re on a mission to end the headaches of ordering synthetic DNA and give researchers what they really want: reliable DNA, on time, exactly as ordered. Synthetic DNA powers almost everything in biological research, whether you’re running thousands of tests with your favorite AI model, screening proteins like there’s no tomorrow, or designing entire genomes from scratch. But the traditional way of building synthetic DNA is like trying to run a marathon … in quicksand. It’s slow, messy, and holds you back. In a world where DNA is the foundation of discovery, these limitations are costing us breakthroughs in new cures, bio-manufactured chemicals, gene editing, and so much more.

Ansa has re-imagined the entire DNA synthesis process. Today, we were thrilled to announce a major leap forward with the expansion of Ansa’s clonal gene offering, which now supports sequences up to 5 kb. This means you can now get your hands on the long, complex DNA you need without the fuss of piecing together smaller DNA pieces or running into continuous delays with unreliable vendors. Just send us your wish list of genes, promoters, terminators, and any other goodies, and we’ll deliver them fully assembled and ready to go.

The word “complexity” is often used by DNA providers to cover up their technology’s limitations. It’s a catch-all term used to justify delays, redesigns, and price hikes. That’s why Ansa is embracing complexity, offering scientists an entirely different experience with ordering DNA. This launch of 5 kb clonal DNA means you can be less stressed about rejected sequences. Hairpins? High GC content? Repeats? We are working to make all that DNA drama is about to be a thing of the past. No more of those seemingly endless back-and-forth redesigns, or those dreaded emails that say your sequence isn’t working. No more doing your own cloning to compensate for a vendor’s limitations.

Ansa’s enzymatic DNA synthesis technology, which is based on a groundbreaking technique for synthesizing oligonucleotides using a TdT enzyme tethered to a single nucleotide, has quickly become the world’s most accurate oligo synthesizer. We produce 600-mer starting oligos that are several times longer than what traditional methods can pull off. By printing genes directly, we bypass one of the most failure-prone steps in gene synthesis — polymerase chain assembly, a process that struggles with high GC content, hairpins, and repetitive sequences.

That’s the core technology that allowed us to launch our 600-mer gene fragment and clonal DNA products earlier this year. But we knew customers needed ready-to-use materials that minimize or even eliminate in-house cloning, so our products had to get much longer. To accomplish this, we needed a revolutionary assembly technique, one that is agnostic to sequence complexity. Our novel, state-of-the-art assembly technology works reliably, regardless of sequence length or complexity. When you pair it with our oligo synthesis platform, you get what we think is the most reliable and scalable DNA synthesis experience out there.

Thanks to this approach, Ansa has already delivered DNA sequences that customers couldn’t get anywhere else. For one client, we synthesized highly repetitive sequences for silk protein research. For another, we built sequences for phage genomes with high GC content. We’ve even synthesized entire viral vectors with multiple inverted terminal repeats and complex cargoes. Time and time again, we’ve heard from customers who had waited months or more for other vendors to deliver, only to be turned away or left in limbo. We’ve turned what was once impossible into just another order fulfilled.

The launch of our 5 kb clonal service is about to shake things up. We’re clearing the path for scientists to dream bigger, work faster, and get more done. And we’ll keep pushing the boundaries to deliver even better products in the future. In the meantime, we are really excited to see what our customers will accomplish now that ordering synthetic DNA just got a whole lot easier.
